Summary:Korean mint [Agastache rugosa (Fisch. & C.A.Mey.) Kuntze] and its bioactive compound tilianin confers various biological functions, including anti-inflammatory, antioxidant, and antiobesity activities. This study investigated the ameliorative effect of A. rugosa extract (ARE) and tilianin on immobilization-induced muscle atrophy in C57BL/6J mice with particular emphasis on the signaling pathways of protein metabolism. Oral administration of 100 and 250 mg/kg/day ARE and 10 mg/kg/day tilianin improved muscle weight, muscle volume, myofiber cross-sectional area, grip strength, and exercise endurance. ARE and tilianin enhanced protein synthesis by stimulating the phosphatidylinositol 3-kinase/Akt/ mammalian target of rapamycin pathway and inhibited protein degradation by suppressing muscle RING-finger protein 1, atrogin-1, and forkhead box O3 translocation. Additionally, ARE and tilianin reduced the expression of inflammatory markers and enhanced the expression of antioxidant enzymes. These results suggest that ARE and tilianin have potential as inhibitory agents against muscle atrophy. KCI Citation Count: 0
