Summary: | In a s ries of comparative studies on virus-host cell interaction of poxviruses, cytochemical, autoradiographic and immunocytological analysis in orf (contagious pustular dermatitis) virus monkey kidney (MK) or bovine kidney (BK) cell system were performed. Secondary or third subcultured monolayer cells of African green monkey kidney or bovine kidney were inoculated with Iwate BT-9 strain of orf virus. Characteristic cytopathic effects (CPE) which consist of rounded and refractile cells were appeared on 4-5 days postinoculation in cultured monolayers of both animal origins. By the proceeded infection there was a general degeneration of the monolayer, and it occurred faster than that of cowpoxvirus-FL cell system. In the stained preparations of infected cells of both kinds, compact or diffuse intracytoplasmic B-type inclusions were observed. The infected MK cells had a tendency to yield rather defined compact inclusions than in the infected BK cells. In the progressed cultures the inclusions became likely to the less defined unclear features. No intracytoplasmic inclusion which correspond to the type-A inclusion was observed in both kinds of cultures, which occurs in cowpoxvirus infected cells. The appearance of silver grains in autoradiograms of the infected monolayers of both origins was coincided with these inclusions, thus it was concluded that these inclusions must be the site of viral DNA synthesis and virus maturation. In the prolonged infected cultures especially BK cells, the accumulation of silver grains coincided also sparsely with more diffuse and unnoticeable cytoplasmic inclusions. By the direct immunofluorescent method using heterologous anticowpox fluorescent antibody, the localization of viral antigens was detected in the cytoplasms and some nuclei of the infected monolayer cells of both kinds. It is suggested that cross reaction is existed between orf and cowpox although some non-specific reactions were observed. |
