Summary: | PROBLEM TO BE SOLVED: To obtain the new subject DNA fragment containing a gene corresponding to biosynthesis of biotin derived from microorganisms belonging to the genus Sphingomonas, used for gene technological breeding of microorganisms producing the biotin useful as a food additive, etc. SOLUTION: This DNA fragment is a new DNA fragment containing genes corresponding to a biotin biosynthesis, such as a 7-keto-8-aminopelargonic acid synthase gene, a 7,8-diaminopelargonic acid aminotransferase gene, a desthiobiotin synthase gene, a biotin synthase gene and a gene of an enzyme catalyzing a reaction of a biosynthesis step upstream from Pimelyl Co-A in a biosynthesis rout of the biotin, and is useful for gene technological breeding, etc., of biotin-producing microorganisms. The DNA fragment is obtained by extracting chromosome DNA of Sphingomonas paucimobilis JCM7511, forming a chromosome DNA library by a conventional method, and recovering a DNA from strains capable of being cultured in a medium containing no biotin. |
