The invention provides an aquaculture method of ostrea plicatula. The method comprises the following steps: 1. selecting an aquaculture area according to the conditions of 5-15m water depth during low tide, 40-60cm/s water speed, substrate of sediment or sand, over 50cm transparency, no eddy and no reverse bump flow; 2. selecting a seedling collection ground which locates in the border region between middle and low tidal regions of inner bar with parent oysters and is the sandy mud flat of slower flow rate and flat terrain; 3. arranging a seedling collection support through taking moso bamboo or batten as support bar and putting up a shelf of 1-1.5m wide and 40-50cm away from the bottom; 4. arranging a cultivation stand of 1.5-2m space among shelf pillars and 1-1.5m away from the bottom; 5. horizontally hanging ostrea strings in the direction of following wind with the string spacing of 15-20cm.
