干细胞核心基因NANOG在乳腺癌干细胞及非干细胞中的表达与定位
目的研究干细胞核心基因NANOG在乳腺癌干细胞(BCSCs)和非乳腺癌干细胞(Non-BCSCs)中的表达及定位差异以及BCSCs在MCF-7微球体中的分布情况。方法应用免疫荧光、免疫细胞化学及NANOG与Hoechst33342荧光染料双染色方法检测BCSCs和Non-BCSCs中NANOG的表达及其定位。结果免疫细胞化学及免疫荧光染色显示,MCF-7中存在两种细胞:一种细胞胞体大、细胞质丰富,NANOG主要表达其细胞质中,这部分细胞占绝大多数;另一种细胞胞体小而圆、细胞质少,NANOG主要表达其细胞核中,这部分细胞占少数。NANOG和Hoechst33342免疫荧光双染显示:NANOG表达...

| Summary: | 目的研究干细胞核心基因NANOG在乳腺癌干细胞(BCSCs)和非乳腺癌干细胞(Non-BCSCs)中的表达及定位差异以及BCSCs在MCF-7微球体中的分布情况。方法应用免疫荧光、免疫细胞化学及NANOG与Hoechst33342荧光染料双染色方法检测BCSCs和Non-BCSCs中NANOG的表达及其定位。结果免疫细胞化学及免疫荧光染色显示,MCF-7中存在两种细胞:一种细胞胞体大、细胞质丰富,NANOG主要表达其细胞质中,这部分细胞占绝大多数;另一种细胞胞体小而圆、细胞质少,NANOG主要表达其细胞核中,这部分细胞占少数。NANOG和Hoechst33342免疫荧光双染显示:NANOG表达于Hoechst33342拒染的细胞核中;而在Hoechst33342核染色的细胞中,NANOG主要表达于细胞质中。微球体呈球囊性、中心部呈空腔状,细胞沿着球壁分布,球壁多为1~3层细胞组成;在MCF-7微球体中,BCSCs主要分布在囊性微球体的内侧壁。结论 NANOG在BCSCs中主要表达在细胞核,而在Non-BCSCs中主要表达在细胞质,而BCSCs主要分布在囊性微球体的内侧壁,这可为BCSCs的进一步研究和乳腺癌的临床靶向治疗提供实验依据。 |

| Bibliography: | Objective To explore the expression and location differences of core gene NANOG of stem cells in breast cancer stem cells(BCSCs) and non-breast cancer stem cells(Non-BCSCs) as well as the distribution of BCSCs in mammospheres of MCF-7.Methods The expression and localization of NANOG in BCSCs and Non-BCSCs were detected by immunohistochemistry,immunocytochemistry,and double staining of NANOG and flourescent dye Hoechst 33342.Results Immunocytochemistry and immunofluorescence staining showed that there were two types of cells in MCF-7.Most cells had big bodies and abundant cytoplasm,and NANOG mainly expressed in the cytoplasm.Other cells had small round bodies and less cytoplasm,and NANOG mainly expressed in the nucleus.NANOG and Hoechst 33342 immunofluorescence double staining showed that NANOG expressed in the nucleus that could not be stained by Hoechst 33342.In cells that the nucleus was stained by Hoechst 33342,NANOG mainly expressed in the cytoplasm.The mammospheres were cystic sphere and hollow in center |
