精神分裂症患者血浆miRNA表达水平与精神症状的关系研究

Summary:目的 探讨精神分裂症患者血浆中miRNA表达水平与精神症状的关系。方法 连续纳入2012年7月－2013年5月收治的53例未经药物治疗的精神分裂症患者，采用实时荧光定量PCR技术检测血浆中9种miRNA（miR-30e、miR-34a、miR-181b、miR-195、miR-346、miR-432、miR-7、miR-132、miR-212）的表达水平，采用阳性和阴性症状量表（PANSS）评估其精神症状，并对结果进行统计学分析。结果 miR-34a与激活性症状及攻击性症状呈显著负相关（r=–0.345、–0.284，P〈0.05），miR-346与一般精神病理症状呈明显负相关（r=–0.282，P〈0.05）。miR-34a高表达组激活性症状群分和攻击性症状群分显著低于低表达组（P〈0.05），miR-346高表达组反应缺乏症状群分显著低于低表达组（P〈0.05）。miR-34a分别进入以激活性症状群和攻击性症状群为因变量的回归方程，可解释激活性症状变异的10.2%，攻击性症状群变异的6.3%；miR-346进入以一般精神病理症状为因变量的回归方程，可解释一般精神病理变异的6.1%。结论 精神分裂症患者血浆中的miR-34a与激活性症状、攻击性症状有明显关联，miR-346与一般精神病理症状有明显关联，此二者可能在上述精神症状的发生发展过程中起重要作用。
Bibliography:Objective To explore the correlation between the microRNA expression in plasma and psychiatric symptoms in schizophrenia patients. Methods A total of 53 consecutive schizophrenia patients who had not received antipsychotic treatment from July 2012 to May 2013 were enrolled in the present study. The expressions of 9 schizophrenia-associated microRNAs （miR30e, miR-34a, miR-181b, miR-195, miR-346, miR-432, miR-7, miR-132 and miR-212） in plasma were determined by real-time fluorescence quantitative PCR, with the psychiatric symptoms evaluated by Positive and Negative Symptoms Scale （PANSS）, and the results were analyzed statistically. Results The expression level of miRNA-34a was significantly negatively correlated with the active symptoms and aggressive symptoms （r=–0.345, r=–0.284, P〈0.05）, and the expression level of miRNA-346 was significantly negatively correlated with the general psychopathologic symptoms （r=–0.282, P〈0.05）.
