Abstract [はじめに] 歯周病は歯周病原細菌の感染に伴う局所の炎症性骨破壊により, 歯を喪失する疾患である. 歯周病原細菌の内毒素(LPS)や莢膜多糖(CP)がin vitroの実験系において, 骨吸収活性や破骨細胞形成能を有することや, LPSやCPによる骨吸収や破骨細胞形成に, プロスタグランディンE2やインターロイキン-1 (IL-1)が関与していることが報告されている. 一方で, IL-1は骨芽細胞とのRANKL-RANKの接触を介することなく, 直接破骨細胞を分化・活性化することが報告されており, 歯周病の発症や進行にIL-1が深く関わると考えられている. IL-1ファミリーには, 代表的なアゴニストであるIL-1α, IL-1βや, アンタゴニストとしてのIL-1レセプターアンタゴニスト(IL-1Ra)などが含まれる. IL-1αとIL-1βはT細胞, B細胞の増殖促進, サイトカインの産生誘導, MMP産生, 破骨細胞の分化・活性化を有し, 細胞内シグナル伝達を引き起こすリガンドである.
