黄芪神仙定年方对辐射小鼠肝氧化应激损伤的防护作用

目的探讨黄芪神仙定年方对X射线辐射后小鼠肝氧化应激损伤的防护作用及可能机制。方法将6~8周龄昆明种小鼠随机分为空白组(NC组)、模型组(IO组)、阳性对照组(TC组)及黄芪神仙定年方低(LH组,4.8mg/kg)、中(MH组,9.6mg/kg)、高(HH组,19.2mg/kg)剂量组,每组10只,分别给予生理盐水、生理盐水、胸腺肽及不同浓度黄芪神仙定年方水煎剂连续灌胃10d,然后采用X射线8Gy全身单次照射,照射后继续灌胃3d,称小鼠体重,观察饮食、饮水量及一般状况。处死小鼠取肝脏,计算肝脏指数;观察肝脏组织形态学变化;测定肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶...

Summary:目的探讨黄芪神仙定年方对X射线辐射后小鼠肝氧化应激损伤的防护作用及可能机制。方法将6~8周龄昆明种小鼠随机分为空白组(NC组)、模型组(IO组)、阳性对照组(TC组)及黄芪神仙定年方低(LH组,4.8mg/kg)、中(MH组,9.6mg/kg)、高(HH组,19.2mg/kg)剂量组,每组10只,分别给予生理盐水、生理盐水、胸腺肽及不同浓度黄芪神仙定年方水煎剂连续灌胃10d,然后采用X射线8Gy全身单次照射,照射后继续灌胃3d,称小鼠体重,观察饮食、饮水量及一般状况。处死小鼠取肝脏,计算肝脏指数;观察肝脏组织形态学变化;测定肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)等氧化应激指标及Nrf2蛋白表达情况。结果与NC组比较,IO组小鼠体重明显降低(P〈0.01),病理切片示小鼠肝细胞排列紊乱,中央静脉严重淤血阻塞,其周围肝细胞变性、坏死,肝组织SOD、GSH-Px活性降低,MDA含量升高(P〈0.05),Nrf2蛋白表达量明显升高(P〈0.01)。与IO组比较,TC组及HH组小鼠体重明显增加(P〈0.01),病理切片示肝组织结构基本正常,肝细胞以中央静脉为中心呈放射状排列,肝组织SOD、GSH-Px活性升高,MDA含量降低(P〈0.05),Nrf2蛋白表达量明显降低(P〈0.01)。结论黄芪神仙定年方对辐射后小鼠肝脏氧化应激损伤具有一定保护作用,其机制可能与抗氧化作用有关。
