钴-60伽玛射线诱导淋巴细胞γH2AX表达的研究
采用流式细胞术检测^60Co γ-射线照射后7H2AX表达的量效和时程关系,同时探讨紫外线联合^60Co γ-射线照射后7H2AX的表达情况。结果表明,不同剂量^60Co γ-射线照射后7H2AX表达量与照射剂量之间呈现明显的剂量效应关系。4Gy ^60Co γ-射线照射后,7H2AX的表达在1h达到峰值,随后逐渐减少。与假照组相比,单纯紫外线照射组7H2AX表达增加,一直稳定维持约6h左右,随后开始减少。紫外线联合^60Co γ-射线照射组与单纯^60Co γ-射线照射比较,γH2AX的表达无显著性差异。结果提示采用流式细胞术检测7H2AX的表达估算受照剂量具有可行性,该法有望成为估算辐射剂...

| Bibliography: | Flow cytometry analysis method was used to detect the changes of yH2AX protein expression in human peripheral blood lymphocytes. In the dose-effect study, peripheral blood was irradiated by 60Co γ-rays with the dose of 0-6 Gy and cultivated for 1 h. Peripheral blood were cultivated for 0-24 h after 4 Gy 60Co γ-rays irradiation for the time-effect study. Then, the peripheral blood was divided into four treatment groups, i.e. UV irradiation, 60Co γ-rays irradiation, UV plus 60Co γ-rays irradiation and control group to detect the changes of protein expression in different time. The results showed that the yH2AX protein expression was in a dose-dependent form. The expression of yH2AX peaked at 1 h after 4 Gy irradiation and began to decrease quickly. The expression of yH2AX in UV irradiation group increased compared with that of the sham-irradiation control group, and the expression of yH2AX peaked at 6 h.
