高效液相色谱法测定榴莲中的姜黄素类化合物
目的建立高效液相色谱法测定榴莲中姜黄素、脱甲氧基姜黄素和双脱甲氧基姜黄素3种姜黄素类物质的含量的分析方法。方法样品经甲醇超声提取,以0.4%乙酸水溶液-乙腈为流动相进行等度洗脱,经TSKgel ODS柱进行分离,采用二极管阵列检测器在425 nm处检测,以外标法定量。结果姜黄素类化合物在2.5~200 ng/m L的浓度范围内线性关系良好,相关系数r〉0.99。姜黄素类化合物的检测限均为10μg/kg,在低、中、高3个加标水平下,榴莲肉中化合物回收率均大于70%,相对标准偏差均低于10%(n=6)。结论该方法快速准确,可适用于榴莲中姜黄素类化合物的检测。...

| Bibliography: | Objective To establish a method for determination of 3 kinds of curcuminoids in durian by high performance liquid chromatography (HPLC), including curcumin, desmethoxycurcumin and bisdemethoxycurcumin. MethodsSamples were extracted by methanol with ultrasonic, separated by a TSKgel ODS column with 0.4% acetic acid water-acetonitrile as mobile phase for isocratic elution, and detected by diode array detector at 425 nm with external standard method for quantification.ResultsCurcuminoids had good linear relationship in the range of 2.5~200 ng/mL with correlation coefficient larger than 0.99. The limits of detection for curcuminoids were all 10μg/kg. The average recoveries spiked at low, middle and high levels were greater than 70% and relative standard deviations were below 10% (n=6).Conclusion This method is rapid, accurate and is suitable for the determination of curcuminoids in durian.
