掌叶半夏提取物PE联合顺铂对宫颈癌荷瘤裸鼠的体内毒性评价

Summary: | 目的 探索掌叶半夏提取物(extract from Pinellia pedatisecta Schott, PE)体内联合顺铂(Cis-dichlorodiammine platinum-II, CDDP)抑制宫颈癌时对裸鼠的毒性,评价PE作为宫颈癌顺铂化疗协同药物的安全性。方法 取40只BALB/c雌性裸鼠,建立CaSki皮下移植瘤模型后,随机分为4组:(1)对照组;(2)PE组:每日20 mg/kg (灌胃);(3)CDDP组:每周5 mg/kg(腹腔注射);(4)PE+CDDP组:PE每日20 mg/kg(灌胃)+ CDDP每周5 mg/kg(腹腔注射)。连续给药3周,观察各组小鼠每日摄食和一般情况,每3天称体质量;第22天采血后处死裸鼠,获取裸鼠重要脏器,测量脏器质量,计算脏器系数;HE染色观察主要脏器组织形态学结构,ELISA检测血清谷丙转氨酶(alanine aminotransferase, ALT)、谷草转氨酶(aspartate aminotransferase, AST)、血尿素氮(blood urea nitrogen, BUN)和血肌酐(creatinine, Cr)水平。结果 给药3周后,各组裸鼠生长良好、饮食正常、皮毛光滑、日常活动活跃,无一只死亡。CDDP 组小鼠体重逐渐下降,实验结束时PE+CDDP组小鼠体重大于CDDP组,差异有统计学意义(P<0.05)。光镜下,各组小鼠心、肝脾、肺、肾、卵巢、子宫组织结构均未见明显异常。血清AST和Cr水平在4组间差异均无统计学意义(P〉0.05)。而CDDP组ALT水平明显高于对照组、PE组和PE+CDDP组小鼠,差异具有统计学意义(P<0.05)。结论 PE能改善CDDP引起的裸鼠体重下降及肝酶升高的作用,是一种安全的中药。 |
