利用连接介导PCR检测外源基因整合位点体系的建立

黑素皮质素受体4(Melanocortin receptor4,MC4R)基因是影响猪生长肥育性能的主效基因之一,是参与调控体重、采食和能量平衡的关键信号物质。文章以转猪MC4R基因阳性小鼠基因组为试验材料,通过连接介导PcR(ligation—mediated PCR,LM-PCR)方法,检测猪Mc4R基因在小鼠基因组上的整合位点。结果表明,通过显微注射导入的猪MC4R基因随机整合到了小鼠6号染色体的37236754位点处。本研究初步建立了利用LM—PCR法对外源基因整合位点检测的体系,可为今后转基因动物检测奠定基础。...

MelanocorUn receptor 4(MC4R) gene is one of the major genes that affect the growing-finishing performance in pigs and is the key signal substances involved in body weight regulation, feed intake and energy balance. This study was designed to detect the integrated porcine MC4R sequence in the positive transgenic mouse genome using ligation-mediated PCR (LM-PCR) method which was saved in our laboratory. The results showed that the porcine MC4R gene randomly integrated into the mouse chromosome 6 37 236 754 site. This study preliminarily established the exogenous gene integration site detection system by using the LM-PCR method and laid the foundation for transgenic animals' detection in the future.
