与肝癌肝移植术后复发相关的肿瘤组织标记物筛选

Summary:目的在肝肿瘤细胞中筛选与肝癌肝移植术后复发相关的差异蛋白,为预测患者预后寻找更灵敏、特异的生物标记物。方法19例符合“上海标准”的肝癌肝移植患者纳入本项研究,6例术后出现肿瘤复发和转移(复发组),其余13例患者均无瘤存活(无瘤生存组)。利用表面加强激光解吸电离一飞行时间质谱技术(surface enhanced laser desorption/ionization-time of flight-mass spectroscopy, SELDI-TOF-MS)建立肝肿瘤细胞蛋白质指纹图谱,生物信息软件(Biomarker Wizard)比较两组之间的蛋白质差异。结果利用弱阳离子交换(weakcation exchange,WCX2)蛋白质芯片在质核比(m/z)2000-30000范围内共检测出163个蛋白峰。在建立的蛋白指纹图谱中,复发和无瘤生存组相比较,6个蛋白差异有统计学意义(P〈0.05)。复发组中上调蛋白4个,m/z分别为2129、2203、2950和3062;下调2个,m/z分别为3708和11856。结论由SELDI-TOF筛选出的肿瘤细胞差异蛋白对判断肝癌肝移植患者预后可能有重要意义,差异蛋白可能与微血管癌栓形成有关。

Objective To screen differential proteins associated with recurrence from tumor tissue in patients undergoing liver transplantation for hepatocellular carcinoma (HCC), and to supply the research basis for searching more sensitive and specific biomarkers involved in predicting prognosis.
