SOCS1基因沉默对人膀胱癌T24细胞增殖、侵袭及化疗敏感性的影响
目的探讨细胞因子信号转导抑制因子1(SOCS1)基因沉默对人膀胱癌细胞增殖、侵袭及化疗敏感性的影响。方法将培养好的人膀胱癌T24细胞随机分为三组,用Lipo2000将重组质粒p U-SOCS1-siRNA(沉默SOCS1基因)、阴性对照质粒分别转染至干扰组、对照组,空白组不做任何处理。基因沉默后,用RT-PCR技术检测细胞中SOCS1 mRNA表达;Western blotting法检测细胞中SOCS1蛋白表达;MTT法检测细胞增殖率;流式细胞术检测细胞周期及化疗药物诱导的细胞凋亡率;Transwell小室模型检测细胞侵袭能力;MTT法检测细胞化疗敏感性。结果与其他两组比较,干扰组SOCS1基...

Methods Human bladder cancer cell line T24 cells were randomly divided into three groups: the interference group( which was transfected by recombinant plasmid p U-SOCS1-siRNA),control group( which was transfected by negative control plasmid) and blank group without any treatment. RT-PCR and Western blotting were used to detect the SOCS1 mRNA and protein expression in human bladder cancer cell line( T24) after transfection. The cell proliferation rate was detected by MTT assay,flow cytometry was used to detect the cell cycle and cell apoptosis rate; the invasive ability of T24 cells in vitro was evaluated by Transwell migration assay and the change of chemosensitivity was determined by MTT.
