鸡DLK1基因的生物信息学分析

DLK1基因是哺乳动物的一个印记基因,DLK1蛋白促进哺乳动物肌肉的生长发育,但抑制脂肪的生长发育。禽类不存在基因组印记现象,目前禽类DLK1基因的功能和调控机制还不清楚。本研究针对鸡等13种动物的DLK1蛋白进行了多序列比较分析、分子进化分析及糖基化分析,同时还比较了人、鼠及鸡的DLK1基因结构、基因同线性、启动子结构以及3′端非编码区结构。结果表明,尽管鸡和人DLK1基因的进化关系较远,但从mRNA拼接形式、基因结构、启动子结构等来看,与鼠相比,鸡和人的DLK1基因有较多相似之处。研究结果为下一步鸡DLK1基因的功能和调控研究提供了参考依据,同时,也为印迹基因的进化研究以及鸡的脂肪生长发育...

DLK1 is encoded by the imprinted gene DLK1 in mammals.DLK1 promotes muscule development,but inhibits adipose development.Imprinted genes do not exist in avian species.The role and molecular mechanism of DLK1 in avian species are still unknown.In current research,multiple sequence alignment of DLK1 protein sequences from 13 different animals,incuding chicken,red jungle fowl,Taeniopygia guttata and turkey,was done using Clustalw 2.0,and phylogenetic tree was established using MEGA4.1 software.Glycosylation sites and typical domains of DLK1 were analyzed using ELM database and SMART database,respectively.Gene structure,gene synteny,promoter and 3′UTR of DLK1 gene,were compared among human,mouse and chicken.Our results suggest that,although the evolution relationship is quite distant between human,mouse,and chicken,chicken is closer to human than mouse in mRNA splicing,promoter and gene structure of DLK1.The data pay the way for future experimental study on DLK1 gene in chicken.
