经大鼠脾动脉行肝细胞移植建立动物模型
目的建立经SD大鼠颈动脉途径脾动脉插管行肝细胞移植的动物模型,并探讨其临床意义。方法采用I)_氨基半乳糖(D-galactosamine,D-gal)诱导大鼠急性肝损伤(acuteliveriNury,ALD,24h后40只SD大鼠分为两组;A组(n=20)经左侧颈动脉插管,进入腹腔干,通过脾动脉移植1×10^7个肝细胞;B组(n=20)通过同样的方法经脾动脉注射0.5mLHank’s液作为对照,观察不同时间点(1、3、6、10、15天)大鼠肝功能的变化情况以及脾内白蛋白合成作用,HE染色观察脾内肝细胞分布情况。结果经颈动脉途径脾动脉插管成功率约90%,荧光示踪剂CFDASE标记的肝细胞脾内移...

Bibliography: | Objective To establish an animal model of hepatocyte transplantation via splenic artery in SD rats,and to discuss it's clinic value. Methods Acute liver injury (ALI) was induced by D-gal in 40 SD rats,and they were randomly derided into group A and group B 24 hours later. In group A (n=20), a catheter was inserted into rats' splenic artery via left common carotid artery, then i x I07 hepatocytes were transplanted into the spleen through the catheter. In group B (n = 20) ,0.5 mL Hank's solution was infused into the spleen as control through the catheter which inserted into rats' splenic artery by the same way as group A. At different time points (1,3,6,10, and 15 days), liver function was measured, and secretion of albumin in transplanted hepatocytes was detected by immunofluorescence, and distribution of hepatocyte in spleen was observed by HE stain.
