新型化合物DJ5对单侧输尿管梗阻小鼠肾纤维化作用的实验研究

Summary:目的:观察新型化合物DJ5对单侧输尿管梗阻(unilateral ureteral obstruction,UUO)小鼠肾功能、肾脏病理和纤维化指标的影响,探讨其对肾纤维化的作用。方法:54只雄性C57BL/6小鼠随机分为9组:6个不同剂量DJ5治疗组、奥美沙坦酯组、UUO模型组及假手术组(n=6)。除假手术组外,其余各组小鼠均行单侧输尿管结扎术,建立UUO小鼠模型。给予各组UUO模型小鼠不同剂量的DJ5(1、3、10、30、60、100mg/kg)、奥美沙坦酯(6.25mg/kg)或甲基纤维素治疗。观察各组小鼠的肾功能、肾脏病理和纤维化指标的变化。应用荧光定量RT-PCR法和蛋白质印迹法检测肿瘤坏死因子β(TGF-β)、纤维连接蛋白(FN)、结缔组织生长因子(CTGF)、α-平滑肌肌动蛋白(α-SMA)、Ⅰ型和Ⅲ型胶原(ColⅠ、ColⅢ)的变化。结果:各剂量DJ5治疗组与模型组比较,肾功能和肾纤维化的差异不明显;TGF-β、FN、CTGF、α-SMA、ColⅠ、ColⅢ与假手术组比较,各手术组均出现肾纤维化;但各剂量DJ5治疗组与模型组比较,TGF-β、CTGF、α-SMA、ColⅠ、ColⅢ无显著差异;DJ5 60和100mg/kg治疗组FN表达较模型组显著减少(P〈0.05)。结论:新型化合物DJ5对肾纤维化的作用有限,仅在高剂量时对FN有显著作用。

Objective:To explore the effects of a new compound DJ5 on renal function,renal pathology and fibrosis-related proteins of unilateral ureteral obstruction(UUO)in the mouse model and investigate its effects on renal fibrosis.Methods:Fiftyfour C57BL/6male mice were randomly divided into 9groups,including 6groups with different doses of DJ5,the olmesartan medoxomil group,the UUO model group and the sham operation group(n=6).Mice in all the groups,except the sham operation group,underwent unilateral ureteral ligature to establish the UUO model.Then the mice in different UUO model groups were given different doses of DJ5(1,3,10,30,60,100mg/kg),olmesartan medoxomil(6.25mg/kg)and methyl cellulose,respectively.Changes in renal functions,renal pathology and fibrosis-related proteins were closely observed in all the groups..Tumor necrosis factorβ(TGF-β),fibronection(FN),connective tissue growth factor(CTGF),α-smooth muscle actin(α-SMA),collagen typeⅠ(ColⅠ)and collagen
