虫生广布拟盘多毛孢蛋白酶、几丁质酶、脂肪酶产生水平及其毒力关系研究

[目的]了解虫生广布拟盘多毛孢GXSTYJ03菌株侵染和杀死松突园蚧过程中酶活性变化及作用机理.[方法]通过Folin-酚法、DNS法和对硝基苯酚法分别测定含1×104、1×105、1×106、1×107和1×108个孢子/mL系列浓度的GXSTYJ03菌株孢子悬浮液产生的蛋白酶、几丁质酶和脂肪酶活力及其对松突圆蚧的致死率.[结果]GXSTYJ03菌株孢子含量越高,其产蛋白酶、几丁质酶、脂肪酶的活力也越高;蛋白酶、几丁质酶、脂肪酶的产酶高峰期和产酶活性均不相同,蛋白酶的产酶高峰期出现在发酵后第5d、几丁质酶出现在发酵后第6d、脂肪酶出现在发酵后第4~5d,GXSTYJ03菌株的蛋白酶活性远大于...

[Objective]The activities of protease,chitinase and lipase produced by entomogenous Pestalotiopsis disseminata GXSTYJ03 were studied to analyze their relationships with virulence.[Method]By using Folin-phenol,DNS and Nitrophenol method,the activities of protease,chitinase and lipase produced by different spore content of entomogenous Pestalotiopsis disseminata GXSTYJ03 such as 1 ×104,1 ×105,1 ×106,1×107 and 1 × 108 spores were measured separately,and the death rate of Hemiberlesia pitysophila Takagi infested by different concentrations of entomogenous Pestalotiopsis disseminata was calculated.[Result]The results showed that the higher concentration of entomogenous Pestalotiopsis disseminata GXSTYJ03 spore,the greater activities of protease,chitinase and lipase.The peak of protease enzyme production was on Day 5 after fermentation,the peak of chitinase enzyme production was on Day 6,and the peak of lipase enzyme production was on Day 4-5.The activity of protease was much larger than that of chitinase
