FSTL1及MMP-2在子痫前期孕妇胎盘组织及滋养细胞中的表达及相关性研究

Summary:目的:探讨卵泡抑素样蛋白1(FSTL1)及基质金属蛋白酶2(MMP-2)在子痫前期孕妇胎盘组织及滋养细胞中的表达及相关性。方法以2014年1月至2015年9月在重庆医科大学附属第一医院产科住院行剖宫产术的子痫前期孕妇35例和正常足月妊娠行择期剖宫产术孕妇40例为研究对象,采用蛋白质印迹法检测正常晚期妊娠孕妇和子痫前期患者胎盘组织中FSTL1和MMP-2蛋白的表达;采用免疫荧光法检测正常培养和缺氧/复氧(H/R)处理的人早孕期绒毛膜外滋养细胞株HTR8/SVneo中FSTL1和MMP-2的表达;采用酶联免疫吸附试验(ELISA)检测两组孕妇血清FSTL1水平,分析FSTL1和MMP-2表达水平与子痫前期的相关性。结果与正常足月妊娠胎盘组织比较,子痫前期患者胎盘组织中FSTL1水平明显升高,而MMP-2水平明显下降,差异均有统计学意义(P<0.05)。与正常培养的细胞比较,H/R处理的细胞FSTL1蛋白水平明显升高,而MMP-2水平明显下降,差异均有统计学意义(P<0.05);FSTL1主要定位于HTR8/SV-neo细胞质中。正常足月妊娠孕妇血清FSTL1水平显著低于子痫前期孕妇,差异有统计学意义(P<0.01)。结论胎盘组织和血清中FSTL1和MMP-2水平的异常变化可能与子痫前期发病有关,二者可能存在协同作用。通过研究FSTL1和MMP-2的相互作用,可为子痫前期发病机制的研究提供新思路。
