3个广藿香品种染色体制片技术优化与计数
以广藿香根尖为试验材料,采用常规压片法比较取材时间、预处理液、处理时间、酶液浓度、酶解时间、低渗时间对广藿香染色体制片效果的影响,采用去壁低渗法对石牌、海南、高要3个广藿香品种进行染色体计数。结果表明:上午10:30-11:00取材、冰水混合液处理24h、4%混合酶液(纤维素酶和果胶酶各占4%,g/V)酶解30min、低渗15min所得广藿香染色体制片效果较好;3个广藿香品种分散良好、清晰的50个分裂相细胞,70%以上的细胞染色体数目为2n=64,3个广藿香品种在染色体数目上没有差异。...
