Summary:Two new flavonoids, 8,2′-diprenyl-3,5,7,5′-tetrahydroxyflavone ( 1 ) and 8-(1-methoxy-2,3-dihydroxy-3-methylbutyl)naringenin ( 2 ), along with twelve known compounds ( 3–14 ), were isolated from the leaves of Macaranga pustulata . The structures of the new compounds 1 and 2 were established by detailed spectroscopic data analysis and ECD computational methods. The inhibitory effects of compounds 1–6 against NLRP3 inflammasome activation as well as the cytotoxic activities of compounds 1–4 against MCF-7 and HCT-116 cancer cell lines were investigated. Compound 3 showed weak cytotoxicity against HCT-116 (IC 50 = 55.747 ± 3.212 μM).
