电离辐射诱导小鼠骨髓树突状免疫细胞急性凋亡的研究
观察X射线辐射对小鼠骨髓树突状免疫细胞(Dendritic Cells,DCs)的影响。利用X射线对小鼠骨髓树突状免疫细胞进行不同剂量的照射,流式细胞术检测小鼠骨髓DCs的凋亡比例、CCK-8方法检测DCs免疫能力的变化情况,利用蛋白抑制剂对JAK2/STAT3信号转导通路进行抑制,探讨可能的分子机理。与未照射组比较,X射线照射0.5 Gy能够诱导小鼠骨髓DCs抗原递呈和抑制肿瘤细胞活性能力下降,凋亡比例增加,JAK2、STAT和Caspase-3蛋白表达显著增加。X射线能够诱导小鼠骨髓DCs凋亡增加,从而导致细胞免疫能力下降,JAK2/STAT3/Caspase-3通路可能是参与X射线诱导D...

[Purpose] This study aims to observe the effect of irradiation on bone marrow dendritic cells(BMDCs) in mice. [Methods] The BMDCs were irradiated by X-ray to absorbed doses of 0.5 Gy or 1.0 Gy. The apoptosis and immunity ability of DCs were measured by flow cytometry and CCK-8. Protein expressions were determined by Western blot. The JAK2/STAT3 signal transduction pathway was inhibited by protein inhibitors. [Results] The dysfunction of BMDCs induced by X-ray irradiation was observed.
