苦龙胆酯苷拮抗高脂饮食所致的小鼠动脉粥样硬化

Abstract | R54%R966; [目的]探究苦龙胆酯苷(MAG)对高脂饮食(HFD)所致动脉粥样硬化的拮抗作用及其机制.[方法]将30只ApoE-/-小鼠随机分为对照组(正常饮食)、HFD组、HFD+MAG组,每组10只小鼠,其中HFD+MAG组每日予50 mg/kg MAG灌胃,持续12周.生物化学检测仪测定血脂水平;油红O染色观察主动脉斑块沉积情况;苏木精-伊红(HE)染色观察主动脉血管病理形态表现;Mac-3免疫组织化学染色观察主动脉巨噬细胞聚集情况;免疫荧光定位染色观察NOD样受体热蛋白结构域相关蛋白3(NLRP3)表达与分布;Western blot检测主动脉中NLRP3、凋亡相关斑点样蛋白(ASC)和Caspase-1亚基p20表达水平.[结果]HFD+MAG组血清甘油三酯、总胆固醇和低密度脂蛋白胆固醇水平分别降低至HFD组的88.39%、74.85%和64.97%(均P<0.05),高密度脂蛋白胆固醇水平升高至HFD组的142.18%(P<0.05),斑块面积减少至HFD组的45.16%(P<0.05),MAG能改善主动脉病理形态表现,使巨噬细胞沉积减少,斑块内NLRP3阳性巨噬细胞比例降低,主动脉中NLRP3、ASC和Caspase-1亚基p20表达分别降低至HFD组的45.58%、53.28%和42.35%(均P<0.05).[结论]MAG对高脂饮食所致ApoE-/-小鼠动脉粥样硬化有明显的拮抗作用,其机制可能与其抑制斑块内巨噬细胞NLRP3炎症小体的激活有关. |
