본 연구에서는 HPH의 C2C12 근육세포 분화 및 근위축 조절 활성을 평가하였다. Abstract | 본 연구에서는 HPH의 C2C12 근육세포 분화 및 근위축 조절 활성을 평가하였다. HPH의 이러한 활성은 원형 단백질인 HPI의 활성보다 월등히 우수하게 관찰되었다. 따라서 이러한 결과는 HPH가 근육분화 및 근위축에 관여하는 주요 인자들의 단백질 발현을 조절함으로써 myotube 형성을 촉진하고 근위축을 예방할 수 있는 가능성을 보여주며, 향후 동물모델에서의 검증 등 추가적인 연구를 통해 새로운 기능성식품 소재로써의 검증이 필요할 것으로 판단된다. Recently, interest in the nutritional and functional properties of industrial hemp seeds has increased. This study was performed to investigate the effects of hemp seed-derived protein hydrolysates (HPH) on myotube formation and muscle atrophy using C2C12 myoblasts. HPH was prepared by the enzymatic (alcalase+flavourzyme or pepsin+pancreatin) treatment of hemp seed protein isolate (HPI). HPH exhibited a higher degree of hydrolysis when treated with alcalase+flavourzyme rather than pepsin+pancreatin. During the differentiation of C2C12 myoblasts, HPH promoted myotube formation and increased the protein expressions of myogenic factor 5, myogenic differentiation 1 (MyoD), and myosin heavy chain (MyHC). In addition, HPH protected C2C12 myotubes against dexamethasone-induced muscle atrophy by increasing the protein expressions of MyoD and MyHC and reducing the expression of muscle RING finger-1. Furthermore, these effects of HPH were more potent than those of HPI.
