Glycoprotein IIb/IIIa의 억제를 통한 갈색거저리 추출물의 혈소판 기능 및 혈전생성 억제 효과

Abstract | 본 연구에서는 갈색거저리 추출물(TME)이 혈소판응집반응과 인테그린 αIIb/β3에 미치는 영향을 평가하였고, 이와 관련된 신호전달 분자들을 어떻게 조절하는지 규명하고자 하였다. 그 결과 TME는 콜라겐이 유도한 platelet aggregation을 강력하게 억제하였고, 세포 내부 칼슘 동원과 외부 칼슘 유입과 관련된 IP3R의 인산화를 억제하고, TXA2의 합성과 관련된 cPLA2/p38, 그리고 인테그린의 활성과 관련된 PI3K/Akt/GSK-3/SYK/VASP의 인산화를 조절한다는 것을 규명하였다. 따라서 TME는 항혈소판 효과가 있는 천연물 자원으로서 치료 및 예방 약물로서 잠재적 가치가 있다고 여겨진다. Platelets play fundamental roles in hemostasis and thrombosis, and antiplatelet drugs effectively reduce thrombosis in patients with cardiovascular disease. However, various side effects have been reported, and although various antiplatelet drugs have been developed, mortality rates of cardiovascular diseases have not decreased. Therefore, new antiplatelet drugs with fewer side effects are urgently required. This study examined whether Tenebrio molitor extract inhibits platelet aggregation by regulating integrin αIIb/β3 and its associated signaling molecules. Tenebrio molitor extract inhibited αIIb/β3 activation by regulating vasodilator-stimulated phosphoprotein, phosphatidylinositol-3 kinase, Akt, and glycogen synthase kinase-3α/β. In summary, Tenebrio molitor extract exhibited strong antiplatelet effects and appears to be a potential therapeutic for preventing platelet-related thrombosis and cardiovascular disease.
