겹삼잎국화 추출물의 항산화 활성과 페놀산 분석

Abstract 본 연구에서는 건조된 겹삼잎국화의 항산화 활성과 함량이 높은 주요 성분들을 확인하고자 하였다. 에탄올 추출물 조성 비율에 따른 겹삼잎국화의 총 폴리페놀 함량은 95% 에탄올 추출물에서 가장 높은 34 mg GAE/g을 갖는 것으로 확인하였다. 또한 항산화능의 지표인 DPPH와 ABTS 라디칼 소거능 측정 결과 타 연구에서 보고한 대부분의 국화보다 더 높은 활성을 나타내었다. 이와 관련된 활성성분을 확인하기 위하여 LC-QToF/MS를 통해 6개의 주요 성분이 함유되어 있음을 확인하였고, 구조 동정 결과 각각 chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, 3,5-dicaffeoyl quinic acid(isochlorogenic acid A), 3,4-dicaffeoyl quinic acid(isochlorogenic acid B), 4,5-dicaffeoyl quinic acid(isochlorogenic acid C)로 추정되었다. 그리고 겹삼잎국화의 chlorogenic acid 이성질체와 유도체들의 정량 분석을 실시한 결과 chlorogenic acid가 183.91 mg/g으로 가장 높은 함량을 나타내었다. 이러한 chlorogenic acid 이성질체와 유도체들은 항고혈압, 항염증 등 다양한 생리활성이 보고된 바 있기에 겹삼잎국화의 식의약소재로 활용 가능성 제기하는 바이다. Polyphenols are known to be beneficial for humans as antioxidants. Especially, chlorogenic acid (CA) is one of the most available acids among all phenolic compounds. Rudbeckia laciniata var. hortensis is Rudbeckia species characterized by an abundance of CA. In this study, ethanol extract of R. laciniata var. hortensis was found to contain a large amount of polyphenols and have significant radical scavenging activity. The IC50 value for 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity was 0.21 mg/mL while that for 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonate) radical scavenging activity was 0.28 mg/mL. We identified three CA isomers and three CA derivatives in R. laciniata var. hortensis using UPLC-QToF/MS. The three CA isomers were chlorogenic acid, neochlorogenic acid, and cryptochlorogenic acid, while the three CA derivatives were 3,5-dicaffeoyl quinic acid (isochlorogenic acid A), 3,4-dicaffeoyl quinic acid (isochlorogenic acid B), and 4,5-dicaffeoyl quinic acid (isochlorogenic acid C).
