래빗아이 블루베리 ‘Bluegem’ 과피의 프로테옴 분석

Abstract | 본 실험에서 래빗아이 블루베리 ‘Bluegem’의 과피에서 발현되는 프로테옴을 이차원 전기연동으로 분석한 결과 총 810개의단백질들이 확인되었으며, 이 중 182개의 단백질들에 대하여 MALDI-TOF / TOF 분석을 실시하였다. 동정된 단백질의 약 30%는 대사와 에너지 관련 단백질이었다. 대사 관련 단백질 중에는 블루베리에서 다량 함유되어 있는 것으로 알려진 anthocyanin생합성을 조절하는 chalcone synthase, anthocyanidin synthase, flavanone 3-hydroxylase 등이 다수 동정되었다. 에너지 관련단백질 중에는 glucogenesis에서 작용하는 phosphoglycerate mutase, enolase, glyceraldehyde-3-phosphate dehydrogenase,phosphoglycerate kinase, triose phosphate isomerase 등이 동정되었고, 광합성 명반응과 관련해서는 다수의 ATP synthase가동정되었다. 그 외에도 peptidyl prolyl isomerase 등을 포함한 protein fate와 heat shock protein 70등을 포함한 cell rescue,defense와 virulence 관련 단백질 등 다양한 단백질들이 존재하는 것으로 확인 되었다. 한편, 분석된 단백질 spot 중 40%에 달하는 74개는 알려지지 않았거나 미분류된 단백질로 동정되었으며, 이에 대한 추가적인 분석이 필요한 것으로 사료되었다. The blueberry is a bush fruit belonging to the family Ericaceae, in the genus Vaccinium. This fruit has a variety of functional substances and has gained popularity recently due to its antioxidative property and various phytochemical contents including potential role in certain diseases like diabetes. Although a proteomic study has been conducted on a variety of fruits, research on the blueberry is still incomplete. In this study, the proteome of the skin of the rabbiteye blueberry, ‘Bluegem’, was analyzed using two-dimensional gel electrophoresis and then identified using a MALDI-TOF / TOF mass spectrometer. Approximately 810 protein spots were detected on 2DE maps among which 182 spots were thereafter identified using mass spectrometry. The functions of the identified proteins were classified according to the GO-MIPS funcat conversion table. As a result, 30% identified protein spots were functionally classified into metabolism-related proteins (glutamine synthetase, chalcone synthase, anthocyanidin synthase, and flavanone 3-hydroxylase) and 30% to energy-related proteins (enolase, malate dehydrogenase, and ATP synthase). Other proteins were associated with protein fate, cell rescue, defense response, and virulence, such as the heat shock protein peptidyl prolyl isomerase. There were 74 unknown or unclassified proteins included in the gel-electrophoresis separation, which accounted for 40% in functional classification.
