Abstract | Many researchers have been actively investigating the issue of stem cell plasticity with conflicting results. With regard to human stem cells, experiments are impeded by the lack of an appropriate assay system and limited information is available at the moment. We utilized our NOD/SCID/γcnull mice model, a highly efficient recipient of human hematopoietic stem cells, to approach this issue. We observed a high percentage of human type hepatocytes developing in the liver-damaged recipient mice after human hematopoietic stem cell transplantation. We are now analyzing the pathway involved in this phenomenon. The NOD/SCID/γcnull mice model provides an excellent tool to explore the human stem cell system in vivo.
幹細胞の可塑性に関して近年活発に研究されているが, いまだに統一的な理解は得られていない.なかでもヒト幹細胞に関する研究は, 実験系の制約から進展が難しい.われわれは, ヒト造血幹細胞移植モデルとしてきわめて生着性の高いNOD/SCID/γcnullマウスを開発し報告してきたが, このマウスに肝障害を与えることで肝臓できわめて高率にヒト型肝細胞が出現することを見いだした.従来のモデルではなしえなかった細胞レベルでの解析が可能となり, 分化過程の解析が進行中である.NOD/SCID/γcnullマウスモデルは造血系のみならず, ヒト幹細胞システムのin vivo解析にすぐれたモデルといえる. |
