Bioartificial endocrine pancreas (diffusion chamber型人工膵島)を構成する選択的透過膜の孔径の検討を行った. Abstract | Bioartificial endocrine pancreas (diffusion chamber型人工膵島)を構成する選択的透過膜の孔径の検討を行った. Diffusion chamberは, 厚さ0.5cm, 直径2.5cmのシリコシリングに選択的透過膜を上下より接着させた太鼓状のもので, その内腔に移植細胞(膵島細胞:MIN6)が封入されている. 選択的透過膜はアクリル製で, それぞれ直径0.2, 0.1および0.05μmの透過孔径を持った膜を用いた. さらに, chamber内に注入する細胞の機能維持を図るために培養基床(剤)としてagaroseや糖鎖を含有しpolymerおよびnicotinamideを用いた. 次に, それぞれの膜孔径を有するdiffusion chamber内MIN6-cellsを封入した後, 糖尿病ラット(streptozotocine-induced diabetic rats)の腹腔内に異種移植し, 血糖調節の可能性を検討した. その結果, diffusion chamber型人工膵島を構成する選択的透過膜の孔径(0.1μm)と移植細胞の機能維持を図る目的で使用した培養基剤との相互作用によって, 免疫抑制剤を使用することなく異種移植が可能であることが示唆された. |
