Vincristine によるラット胎仔の口蓋裂発生に関する走査電子顕微鏡的および光学顕微鏡的観察

口蓋裂の発生機序解明を目的として,妊娠8日12時間のWistar系ラットにvincristine 0.2mg/kgを単一腹腔内投与し,胎齢14日18時間から19日0時間まで6時間ごとの胎仔二次口蓋の走査電顕的および光顕的観察を行い,非投与群の正常胎仔と比較して次のごとき結果を得た. 1)胎齢16目12時間から19日0時間にいたるvincristine投与群生存胎仔の45.5%に口蓋裂が発生した.その多くは正中顔面裂を合併するもので,その他の口蓋裂でも外形異常とくに頭部異常を伴っていることが多かった. 2)投与群の口蓋裂胎仔では口蓋板の著しい低形成,口蓋板の水平転位不全ないし遅延,癒合不全などが...

Abstract 口蓋裂の発生機序解明を目的として,妊娠8日12時間のWistar系ラットにvincristine 0.2mg/kgを単一腹腔内投与し,胎齢14日18時間から19日0時間まで6時間ごとの胎仔二次口蓋の走査電顕的および光顕的観察を行い,非投与群の正常胎仔と比較して次のごとき結果を得た. 1)胎齢16目12時間から19日0時間にいたるvincristine投与群生存胎仔の45.5%に口蓋裂が発生した.その多くは正中顔面裂を合併するもので,その他の口蓋裂でも外形異常とくに頭部異常を伴っていることが多かった. 2)投与群の口蓋裂胎仔では口蓋板の著しい低形成,口蓋板の水平転位不全ないし遅延,癒合不全などがみられた. 3)投与群で水平転位不全を示した胎仔には下顎の低形成を伴うものがあった.また,癒合不全による口蓋裂胎仔の口蓋板内側縁上皮細胞には分化遅延を示す所見が認められた. 4)投与群では蝶筋陥凹やJacobson器官の低形成ないし無形成が観察された. 5)vincristineによる口蓋裂の発生には上記のごとき変化が関与するが,とくに顔面正中部の低形成ないし欠損,鼻上顎体の形成不全に起因することが多いものと考えられた.
