Abstract Analysis of the permeability of complement and immunoglobulin. The faculty of immunoisolation is an indispensable function for a bio-artificial endocrine pancreas using a diffusion chamber. In this study, we investigated the effect of the attachment of protein to the immunoisolating membrane, and the permeability of immunoglobulin and complement through the membrane. The results showed that protein attachment reduced the pore size of the membrane by 25%, but the number of open pores did not decrease. The permeability rates of immunoglobulin and complement though the membrane were 100% after 12-24 hours. However, potentiation of the immunoisolating membrane and the mixed matrix suppressed the permeability rates of immunoglobulin and complement. In conclusion, the Bio-AEP using the diffusion chamber which we developed possesses a superior immunoisolating function. The possibility for clinical application of the Bio-AEP for a long term xenotransplantation without immunosuppressant is indicated. Diffusion chamber型bio-artificial endocrine pancreas (DC型Bio-AEP)において、免疫隔離機能は必要不可欠なものである。今回われわれはin vitroの実験系で、免疫隔離膜への血清蛋白吸着の影響、および免疫グロブリン、補体の膜透過性を検討した。その結果、血清蛋白の吸着により膜の孔径は3/4に縮小したが、その開存数には変化を認めなかった。免疫グロブリンや補体の膜透過性の実験では24時間以内に75~100%の透過性を見たが、mixed matrixを免疫隔離機構としてともに用いた場合、膜とめ相乗作用によってその透過性は抑制され、さらに補体は失活した。このことより、われわれの開発したDC型Bio-AEPは免疫隔離能を備えたものであり、将来における免疫抑制剤を必要としない長期異種移植への臨床応用の可能性を示しているといえよう。
