超高效液相色谱法检测香菇菌多糖片单糖组分

Abstract_FL Objective To establish a pre-column derivatization method for determination of monosaccharide compositions in Lentinus edodes polysaccharide tables by ultra performance liquid chromatography (UPLC).Methods In order to eliminate the interference in the compound of Lentinus edodes polysaccharide tables,the monosaccharide compositions were determined by UPLC.Qualitative analysis was used on the monosaccharide components ofLentinus edodes polysaccharide raw materials and the monosaccharide was analyzed in raw materials.The optimization and methodology of UPLC were investigated.The main monosaccharide components of Lentinus edodes polysaccharide raw materials were analyzed.The proportions of monosaccharide and molar ratios of polysaccharides were determined.Results The monosaccharide compositions of Lentinus edodes polysaccharide tablet were D-mannose,D-glucose and D-galactose by UPLC.The molar ratios of monosaccharides (D-mannose:D-glucose:D-galactose) in Lentinus edodes polysaccharide tablet were 4.6∶4.28∶6.375,respectively.The recoveries of these monosaccharides were 96.6%~103.8% and relative standard deviations were 2.0%~2.6%.The standard curve of each monosaccharide exhibited a good linear correlation with the coefficient higher than 0.999.Conclusion The established method is rapid,efficient and accurate.It also provides the quality control for Lentinus edodes polysaccharide tablet.
