甘薯悬浮细胞系的建立及对抗草甘膦基因的遗传转化

选取8个四川主栽甘薯品种为材料,剥离茎尖诱导胚性愈伤组织,建立胚性细胞悬浮培养系。以烟草叶片和川薯20悬浮细胞为受体材料,利用农杆菌浸染方法转化抗除草剂基因EPSPS。结果表明,经PCR检测及草甘膦喷施试验后,获得了抗草甘膦阳性甘薯植株13株、阳性烟草植株25株,此实验为以EPSPS基因作为遗传转化的标记基因提供了帮助。...

Eight Sichuan's main varieties of sweet potato were used as tested materials,and their embryonic callus induction and suspension cell culture system by stripping stem tip was established,then plant expression vectors containing herbicide resistance gene EPSPS were introduced into tobacco leaves and chuanshu20 suspension cell by agrobacterium-mediated transformation. The result showed that by PCR analysis and herbicide spraying test,13 positive sweet potato plants and 25 positive tobacco plants of herbicide resistance were observed,which provided a help for herbicide resistance EPSPS gene as a marker gene in transformation.
