甘蔗品种RSD感病性检测及RSD病原菌分离培养

为了解不同甘蔗品种的甘蔗宿根矮化病(RSD)感病情况,采用PCR检测方法,对13个甘蔗品种进行RSD感病率检测.结果显示,在所检测的13个甘蔗品种中,不同品种感病率有所差异,其中所检测的2个CP系列感病率最低.同时,利用MSC培养基从感染RSD的甘蔗蔗汁中分离培养到一种生长缓慢、菌落颜色为乳白色的杆状细菌.根据菌落形态、显微镜观察,以及特异引物和16S rDNA扩增多种方法鉴定,最终确认所分离到的杆状细菌即为甘蔗宿根矮化病病原菌(Lei sonia xyli subsp.xyli)....

sugarcane ratoon stunting disease (RSD); infective rate; isolation
In order to understand the infective rate of ratoon stunting disease (RSD)of different sugarcane varieties, 13 sugarcane varieties were identified by PCR. The results showed that the infective rates of different sugarcane varieties were different, and two CP sugarcane varieties had the lowest infective rates. And a kind of coryneform bacterium with slow growth and milky white colony was isolated from RSD infected sugarcane juice by MSC culture. By colonial morphology, microscopic examination, specific primers and identification of 16S rDNA, the coryneform bacterium was identified as Leifsonia xyli subsp, xyli, the pathogen of sugarcane RSD.
