石栗树腋芽外植体消毒方法的筛选

【目的】筛选石栗树腋芽外植体的消毒方法,解决石栗组织培养中大田外植体污染问题,提高石栗组织培养的成活率。【方法】设计3因子3水平的正交试验,3因子分别是自来水冲洗、0.1%HgCl2和10%NaCIO3溶液;3水平是自来水冲洗时间分别为60、120、180min,0.1%HgCl2溶液浸泡时间分别为0、8、16min,10%NaCIO3溶液浸泡时间分别为0、12、24rain。观察9种处理后石栗枝条表面消毒效果。【结果】自来水冲洗120min,0.1%HgCl2溶液浸泡16rain,10%NaCIO3液浸泡24min,可使污染率降到最低,为16.67%,而诱导愈伤组织成功率达N80.00%。【...

[ Objective ]Appropriate sterilization methods for explants of Aleurites moluccana axillary bud was selected in order to solve the pollution problem in tissue culture of Aleurites moluccna and improve survival rate of tissue-cultured Aleurites moluccana. [Method]Three factors (tap water flushing, 0.1% HgC12 soaking, and 10% NaC103 soaking) and three levels of (60,120, and 180 rain for tap water flushing; 0, 8,16 rain for 0.1% HgC12 soaking; 0,12, 24 rain for 10% NaCl03 soaking) orthogonal test was designed to observe the sterilization effect on appearance of Aleudtes moluccana branch processed by nine treatments. [ Result ]The results showed that the best combination for explants sterilization was as follows: flushed by tap water for 120 rain, soaked in 0.1%HgC12 for 16 rain, and soaked in 10%NaC103 for 24 rain.
