南极磷虾油中总砷含量及砷形态分析

| Summary: | 南极磷虾油富含Omega-3磷脂而具有强大的保健功能,但由于其砷含量较高,在保健品领域的应用受到一定限制。由于南极磷虾油中含有大量的磷脂质,采用常规的提取方法分析难以对其砷形态进行准确定量分析。本研究采用高效液相色谱-电感耦合等离子体质谱((HPLC-ICP-MS)联用技术建立了针对砷甜菜碱(As B)、二甲基砷酸(DMA)、一甲基砷酸(MMA)、三价砷As(Ⅲ)、五价砷As(Ⅴ)5种砷形态的分析方法。采用C_(18)固相萃取进行样品脱脂前处理,采用阴离子交换色谱Ion Pac AS9-H阴离子分析柱(250 mm×4 mm),流动相采用38 mmol/L Na_2SO_4+15 mmol/L乙酸钠;流速0.6 m L/min,流速1 m L/min,进行5种形态砷化合物的分析。结果表明,5种砷化合物的线性范围为0.5~500μg/L,相关系数R〉0.9993.检出限(LOD)为0.1~0.2μg/L,定量限(LOQ)为1.5~2.6μg/L,RSD〈5%。加标回收率为88.9%~106.3%。样品的测定结果显示,南极磷虾油中砷形态的主要存在形式为无毒的砷甜菜碱,因此可以安全地应用于食品及保健品。 |
