人单核细胞白血病多器官浸润裸鼠模型的建立

Abstract R-332; 背景与目的:急性白血病常伴有髓外多器官的浸润,髓外复发为白血病患者的一种复发形式,而白血病细胞浸润机制并未完全阐明.本文通过建立人单核细胞白血病裸鼠多器官浸润模型,为对其机制进行研究提供模型基础.方法:对4和6周龄的BALB/c裸鼠进行切脾(splenectomy),环磷酰胺腹腔注射(cytoxan intraperitoneal injection)及全身亚致死剂量辐照(sublethal irradiation)等预处理(SCI)后,经尾静脉接种5×106或1×107个人单核细胞白血病SHI-1细胞,应用RT-PCR、组织病理切片、免疫组织化学以及流式细胞术等检测裸鼠各脏器中SHI-1细胞浸润生长情况.结果:SHI-1细胞可植入经预处理的裸鼠体内,检测发现各组裸鼠全身多脏器如肝、肺、肾、睾丸均可受累,并可于体表及体内多部位形成淡绿色的肿块.4周龄裸鼠可于多部位如胃、头颅、椎体、纵隔内、骨骼上形成肿块,并于骨髓中可检测到人CD45阳性细胞,平均为2.85%;而6周龄组仅于少部分脏器形成肿块,骨髓中不能检测到人CD45阳性细胞.结论:人单核细胞白血病细胞株SHI-1能在经SCI预处理的裸鼠体内形成白血病及多器官浸润模型,可作为研究白血病及髓外浸润的一种有用工具.
